Common use

Afinitor (everolimus) is prescribed for the treatment of advanced cancers, including renal cell carcinoma, breast cancer, neuroendocrine tumors, and subependymal giant cell astrocytoma associated with tuberous sclerosis. It works by inhibiting the mTOR (mammalian target of rapamycin) pathway, reducing cell growth, division, and tumor progression.

Dosage and direction

Afinitor is taken orally, usually once daily, with or without food. Swallow tablets whole with water. Dosage is individualized based on the type of cancer, patient’s health condition, and other medications. Do not crush, chew, or dissolve tablets. Regular monitoring of blood counts, kidney and liver function, and blood glucose is recommended.

Precautions

Inform your doctor if you have liver or kidney problems, infections, diabetes, or a history of lung disease. Avoid exposure to infections, and report fever, cough, or unusual symptoms promptly. Monitor for mouth ulcers, swelling, or skin rashes.

Contraindications

Do not use Afinitor if allergic to everolimus or any component of the tablet. Avoid use during pregnancy unless specifically directed by a healthcare provider.

Possible side effect

Common side effects include mouth sores, fatigue, rash, diarrhea, nausea, and swelling. Serious side effects may include severe infections, liver or kidney dysfunction, pneumonitis, or increased risk of bleeding. Seek medical attention if severe symptoms occur.

Drug interaction

Afinitor may interact with strong CYP3A4 inhibitors or inducers, immunosuppressants, and certain vaccines. Inform your doctor of all prescription drugs, over-the-counter products, and supplements you are taking.

Missed dose

If a dose is missed, take it as soon as you remember. If it is near the time of the next dose, skip the missed one. Do not double the dose.

Overdose

Overdose may cause severe mouth sores, fatigue, infections, or organ dysfunction. Seek immediate medical attention if overdose is suspected.

Storage

Store Afinitor at room temperature, away from moisture, heat, and light. Keep tablets in the original container and out of reach of children.
